ÖZET FARKLI ÇÖZGEN KULLANIMININ HİNT İNCİRİ (Opuntia Ficus-Indica L.) MEYVESİ ÖZÜTLERİNİN BİYOAKTİF ÖZELLİKLERİ ÜZERİNE ETKİSİ Emrah KILIÇ Yüksek Lisans, Gıda Mühendisliği Anabilim Dalı Danışman: Doç. Dr. Aysun ŞENER GEDÜK Eylül 2022, 50 sayfa Bu çalışmada hint inciri (opuntia ficus-indica l.) meyvesinin toplam fenolik madde içeriği, toplam antioksidan aktivitesi, antibakteriyel aktivitesi ile diyabette kilit konumdaki ?-glikozidaz ve ?-amilaz üzerine inhibisyon etkisi incelenmiştir. Metanol, etanol ve hekzan çözgenleri kullanılarak elde edilen özütlerin toplam fenol miktarının gallik asit cinsinden 0,01-0,160 mg/ml arasında değişmiş ve en yüksek toplam fenolik madde metanollü özütte saptanmıştır. Meyve özütlerinin antioksidan kapasiteleri, DPPH yöntemine göre serbest radikal giderici etkisi 0.23-1.19 mg/ml arasında değişirken FRAP yönteminde indirgeme gücü kapasitesi 0.08-1.40 mg/ml arasında değişmiştir. En yüksek antioksidan potansiyel metanollü özütte saptanmıştır. ?-glikozidaz inhibisyonu IC50 cinsinden 336.66-406.06 mg/ml arasında değişiklik gösterirken, ?-amilaz inhibisyon değerinin ise IC50 cinsinden 11.82-34.09 mg/ml arasında değiştiği belirlenmiştir. ?-glikozidaz ve ?-amilaz enzimlerini inhibe etmede etil alkollü özüt en etkili bulunmuş olup değerler sırasıyla 336.66 mg/ml ve 11.82 mg/ml tespit edilmiştir. Hekzan özütünün herhangi bir inhibisyon etkisi olmamıştır. Hint inciri meyve özütlerinde sadece metanol ile elde edilen özüt Escherichia coli ve Staphylococcus aureus'a karşı etki göstermiştir. Değerler sırasıyla 3.26±0.16 mm ve 2.23±0.12 mm düzeylerindedir. Kontrol olarak kullanılan ampisilin değerleri ise sırasıyla 14.2±0.36 mm ve 17.36±0.12 mm düzeylerinde tespit edilmiştir. Sonuç olarak metanol ile elde edilen meyve özütünde antibakteriyel aktivite değerinin kontrol için kullanılan ampisilin değerlerine göre düşük olduğu belirlenmiştir. Prof. Dr. Aysun ŞENER GEDÜK September 2022, 50 pages In this study, bioactive properties of cactus pear (Opuntia ficus-indica L.) fruit such as total phenolic, total antioxidant, inhibition effect on ?-glucosidase and ?-amylase, which are key in diabetes, and antimicrobial activity were investigated. While the total phenol content of the extracts obtained by using methanol, ethyl alcohol and hexane varied between 0.01-0.160 mg/ml equivalent to gallic acid, the highest total phenolic content was observed in the methanolic extract. Antioxidant capacities of fruit extracts, free radical scavenging effect varied between 0.23-1.19 mg/ml according to DPPH method, while reducing power capacity changed between 0.08-1.40 mg/ml in FRAP method. The strongest antioxidant effect was found in methanolic extract. ?-glucosidase inhibition ranged between 336.66-406.06 mg/ml in terms of IC50, while inhibition of ?-amylase ranged between 11.82-34.09 mg/ml in terms of IC50. Ethyl alcohol extract was found to be the most effective in inhibiting ?-glucosidase and ?-amylase, and the values were determined as 336.66 mg/ml and 11.82 mg/ml, respectively. There was no inhibition effect in hexane extracts. In cactus pear fruit extracts, only the extract obtained with methanol showed activity against Escherichia coli and Staphylococcus aureus. The values are 3.26±0.16 mm and 2.23±0.12 mm, respectively. Ampicillin values used as controls were found to be 14.2±0.36 mm and 17.36±0.12 mm, respectively.
